Valaciclovir 500 mg daily appears to reduce the risk of transmission of HSV-2 to a previously uninfected sexual partner.
A topic in Clinical Evidence summarizes the results of one RCT with a total of 1484 serodiscordant couples. Valaciclovir 500 mg once daily taken by the infected partner reduced the risk of HSV-2 transmission at 8 months (1.9% with valaciclovir vs 3.6% with placebo, HR 0.52, 95% CI 0.27 to 0.99) and the risk of symptomatic HSV-2 (0.5% vs 2.2%, HR 0.24, 95% CI 0.08 to 0.75).
Comment: The quality of evidence was downgraded by sparse data.
